Improvement in Physicochemical Properties of Indomethacin by Melt Granulation Technique
ثبت نشده
چکیده
Indomethacin (IM), most widely used non-steroidal anti-inflammatory drug widely used in developing countries, is classified in Class II in the Biopharmaceutics Classification Systems; this means that IM has very low water solubility and high permeability, thus the dissolution is the absorption rate-limiting factor. The aim of this work was to evaluate the suitability of melt granulation technique for enhancing the solubility, dissolution rate and other physicochemical properties of IM.Granules prepared by melt granulation using polyethylene glycol 4000 and poloxamer 188 as meltable binders and microcrystalline cellulose (MCC) and sodium starch glycolate (SSG) as filler and superdisintegrants respectively. The results showed that all the prepared melt granules shows higher solubility and improvement in dissolution rate comparative to pure IM and their physical mixture. The melt granules with MCC and SSG had a significant (** P<0.01) higher solubility dissolution rate in 15 minutes comparative to other granules and pure IM. The melt granule also shows improvement in flowability and wettability. In conclusion, melt granulation could be proposed as solvent free, rapid and low expensive manufacturing methods to increase the solubility and in vitro dissolution rate of IM.

Investigation of Solid Dispersion Technique in Improvement of Physicochemical Characteristics of Ibuprofen Powder
Ibuprofen solid dispersions were prepared by the solvent and fusion-solvent methods using polyethylene glycol (PEG), polyvinylpyrrolidone (PVP), eudragit RS PO, eudragit RL PO and hydroxypropylmethylcellulose (HPMC) as carriers to improve physicochemical characteristics of ibuprofen. The prepared solid dispersions were evaluated for the flowability, solubility characteristics and dissoluti...
متن کاملFine granules showing sustained drug release prepared by high-shear melt granulation using triglycerin full behenate and milled microcrystalline cellulose.
This study aimed to prepare fine granules with a diameter less than 200μm and sustained drug release properties by melt granulation. Triglycerin full behenate (TR-FB) was examined as a new meltable binder (MB) by comparison of its properties with those of glycerin monostearate (GM), widely used as MB. The effect of milling microcrystalline cellulose (MCC), an excipient for melt granulation, on ...
متن کاملEnhancement of solubility and dissolution rate of Fenofibrate by melt granulation technique
This work describes a melt granulation technique to improve the solubility and dissolution characteristics of a poorly water-soluble drug Fenofibrate (FNO). Melt granulation technique is a process by which pharmaceutical powders are efficiently agglomerated by a meltable polymers and surfactants. The advantage of this technique compared to a conventional granulation is that no water or organic ...

The study investigates ibuprofen with wax-like properties as a multifunctional agent (as an active component and as a melt binder). Binding efficiency was compared with granules prepared by wet granulation using polyvinylpyrollidone (PVP K-30) as a binder for micromeritic, physical and mechanical properties such as angle of repose, particle size distribution Carr's index, Hausner's ratio, crush...
متن کاملMelt Granulation in Fluidised Bed: Screening of the Process Variables by Factorial Design
In recent years, the interest in melt granulation has increased due to the advantages of this technique respect to traditional wet granulation; utilizable equipments for melt granulation are high-shear mixer and fluidised bed [1]. The aim of the study was to evaluate, by asymmetrical factorial design [2], the contribution of some variables on the in situ melt granulation process performed in fl...
